\u57fa\u56e0 \u2165 \u578b\u65b0\u57ce\u75ab\u75c5\u6bd2\uff08NDV\uff09\u662f\u76ee\u524d\u9020\u6210\u6211\u56fd\u9e3d\u65b0\u57ce\u75ab\u6d41\u884c\u7684\u4e3b\u8981\u57fa\u56e0\u578b\u3002\u56fd\u5bb6\u65b0\u57ce\u75ab\u53c2\u8003\u5b9e\u9a8c\u5ba4\u76d1\u6d4b\u6570\u636e\u663e\u793a\uff0c\u8fd1\u5e74\u6765\u9e3d\u6e90 NDV \u5206\u79bb\u7387\u5448\u4e0a\u5347\u8d8b\u52bf\u3002\u4e3a\u5b9e\u73b0\u57fa\u56e0 \u2165 \u578b NDV \u5feb\u901f\u68c0\u6d4b\u4e0e\u7cbe\u786e\u5b9a\u91cf\uff0c\u672c\u7814\u7a76\u9488\u5bf9\u6211\u56fd\u6d41\u884c\u57fa\u56e0 \u2165 \u578b NDV \u7684 F<\/em> \u57fa\u56e0\u4fdd\u5b88\u533a\u8bbe\u8ba1\u7279\u5f02\u6027\u5f15\u7269\u4e0e\u63a2\u9488\uff0c\u5efa\u7acb\u53cd\u8f6c\u5f55\u6570\u5b57 PCR\uff08RT-dPCR\uff09\u68c0\u6d4b\u65b9\u6cd5\uff0c\u5e76\u7cfb\u7edf\u8bc4\u4ef7\u8be5\u65b9\u6cd5\u7684\u7075\u654f\u5ea6\u3001\u7279\u5f02\u6027\u53ca\u91cd\u590d\u6027\u3002\u7ed3\u679c\u8868\u660e\uff0c\u6240\u5efa\u7acb\u7684 RT-dPCR \u65b9\u6cd5\u7ebf\u6027\u5173\u7cfb\u826f\u597d\u3001\u7075\u654f\u5ea6\u4f18\u5f02\uff0c\u6700\u4f4e\u68c0\u6d4b\u9650\u4e3a1.97 copies\/\u03bcL\uff1b\u7279\u5f02\u6027\u826f\u597d\uff0c\u4e0e\u5176\u4ed6\u57fa\u56e0\u578b\u65b0\u57ce\u75ab\u75c5\u6bd2\u5f3a\u6bd2\u682a\u53ca\u5e38\u89c1\u79bd\u7c7b\u75c5\u539f\u75c5\u6bd2\u5747\u65e0\u4ea4\u53c9\u53cd\u5e94\uff1b\u91cd\u590d\u6027\u53ef\u9760\uff0c\u53d8\u5f02\u7cfb\u6570\u4e3a2.2 %\u3002\u91c7\u7528\u8be5\u65b9\u6cd5\u5bf9\u4e34\u5e8a\u6536\u96c6\u7684180\u4efd\u9e3d\u53e3\u54bd \/ \u6cc4\u6b96\u8154\u62ed\u5b50\u6837\u54c1\u8fdb\u884c\u68c0\u6d4b\uff0c\u68c0\u6d4b\u7ed3\u679c\u4e0e\u75c5\u6bd2\u5206\u79bb\u7ed3\u679c\u543b\u5408\u3002\u7efc\u4e0a\uff0c\u672c\u7814\u7a76\u5efa\u7acb\u7684 RT-dPCR \u65b9\u6cd5\u7075\u654f\u5ea6\u9ad8\u3001\u7279\u5f02\u6027\u5f3a\u3001\u91cd\u590d\u6027\u4f73\uff0c\u9002\u7528\u4e8e\u57fa\u56e0 \u2165 \u578b NDV \u7684\u5feb\u901f\u7b5b\u67e5\u4e0e\u7cbe\u51c6\u5b9a\u91cf\uff0c\u4e5f\u53ef\u4e3a\u57fa\u56e0 \u2165 \u578b NDV \u6838\u9178\u6807\u51c6\u7269\u8d28\u7684\u7814\u5236\u63d0\u4f9b\u6280\u672f\u652f\u6491\u3002<\/p>\n\n\n\n

Newcastle disease virus (NDV) genotype \u2165 is the predominant genotype responsible for the prevalence of pigeon Newcastle disease (ND) in China. Surveillance data from the National Newcastle Disease Reference Laboratory revealed that the isolation rate of pigeon-origin NDV has increased in recent years. To achieve rapid detection and accurate quantification of NDV genotype \u2165, specific primers and probes were designed targeting the conserved region of the F<\/em> gene of prevalent NDV genotype \u2165 strains, and a reverse transcription digital PCR (RT-dPCR) assay was established. Subsequently, its sensitivity, specificity and repeatability were systematically evaluated. The results showed that this RT-dPCR assay exhibited favorable linearity and high sensitivity, with a limit of detection of 1.97 copies\/\u03bcL. It had strong specificity with no cross-reactivity against virulent strains of other NDV genotypes and common avian pathogenic viruses, and presented good repeatability with a coefficient of variation of 2.2 %. A total of 180 clinical pigeon oropharyngeal and cloacal swab samples were tested, and the detection results were consistent with those obtained by virus isolation. In summary, the established RT-dPCR assay possesses high sensitivity, specificity and repeatability, which is suitable for rapid detection and precise quantification of NDV genotype \u2165, and also provides a basis for the development of nucleic acid reference materials for NDV genotype \u2165.<\/p>\n\n\n\n
