Abstract<\/h2>\n\n\n\n

\u4e3a\u63d0\u9ad8\u725b\u7ed3\u8282\u6027\u76ae\u80a4\u75c5\u75c5\u6bd2\uff08LSDV\uff09\u6838\u9178\u68c0\u6d4b\u7ed3\u679c\u51c6\u786e\u6027\uff0c\u672c\u7814\u7a76\u7ecf\u75c5\u6bd2\u6838\u9178\u63d0\u53d6\u3001\u8d28\u91cf\u68c0\u5b9a\u4e0e\u5206\u88c5\uff0c\u5236\u5907 LSDV NMG \u682a\u57fa\u56e0\u7ec4 DNA \u6807\u51c6\u7269\u8d28\u3002\u91c7\u7528\u8367\u5149\u5b9a\u91cf PCR\uff08qPCR\uff09\u7b5b\u67e5\u5916\u6e90\u75c5\u6bd2\uff0c\u8bc4\u5b9a\u6837\u54c1\u7eaf\u5ea6\uff1b\u8fd0\u7528\u5fae\u6ef4\u5f0f\u6570\u5b57 PCR\uff08ddPCR\uff09\u68c0\u6d4b ORF101 \u57fa\u56e0\u62f7\u8d1d\u6570\uff0c\u5f00\u5c55\u5747\u5300\u6027\u3001\u7a33\u5b9a\u6027\u8003\u5bdf\uff0c\u5e76\u8054\u5408 9 \u5bb6\u5b9e\u9a8c\u5ba4\u5b8c\u6210\u534f\u540c\u5b9a\u503c\uff1b\u540c\u65f6\u501f\u52a9 qPCR \u5f00\u5c55\u5b9e\u9645\u5e94\u7528\u9a8c\u8bc1\u3002\u7ed3\u679c\u8868\u660e\uff0c\u6807\u51c6\u7269\u8d28\u65e0\u5916\u6e90\u75c5\u6bd2\u6c61\u67d3\uff1b\u7ec4\u5185\u3001\u7ec4\u95f4\u68c0\u6d4b\u7ed3\u679c\u65e0\u663e\u8457\u5dee\u5f02\uff0c\u5747\u5300\u6027\u826f\u597d\u3002\u6837\u54c1\u5728 – 20 \u2103\u53ef\u7a33\u5b9a\u4fdd\u5b58 6 \u4e2a\u6708\uff0c4 \u2103\u53ef\u7a33\u5b9a\u5b58\u653e 7 d\uff0c25 \u2103\u6761\u4ef6\u4e0b\u7a33\u5b9a 24 h\u3002\u8be5\u6807\u51c6\u7269\u8d28 ORF101 \u57fa\u56e0\u5b9a\u503c\u4e3a (8.6 \u00b1 1.3) \u00d7 10\u00b3 \u62f7\u8d1d \/\u03bcL\uff0c\u9002\u7528\u4e8e qPCR \u68c0\u6d4b\u3002\u7efc\u4e0a\uff0c\u8be5\u6807\u51c6\u7269\u8d28\u5b9a\u503c\u7cbe\u51c6\u3001\u5747\u5300\u6027\u4f18\u5f02\uff0c\u53ef\u7528\u4e8e LSDV \u6838\u9178\u68c0\u6d4b\u8d28\u91cf\u7ba1\u63a7\u3001\u4eea\u5668\u6821\u51c6\u4e0e\u65b9\u6cd5\u8bc4\u4ef7\uff0c\u4e5f\u53ef\u4e3a\u4f53\u5916\u8bca\u65ad\u8bd5\u5242\u7814\u53d1\u63d0\u4f9b\u57fa\u7840\u6750\u6599\u3002<\/p>\n\n\n\n

In order to improve the accuracy of lumpy skin disease virus (LSDV) nucleic acid detection, genomic DNA reference material (RM) of the LSDV NMG strain was prepared via nucleic acid extraction, quality verification, and aliquoting. Exogenous virus detection was performed to verify the purity of the RM. A droplet digital PCR (ddPCR) method was used to evaluate the homogeneity and stability of the RM, and collaborative quantification was completed by 9 laboratories. The applicability of the RM was validated by quantitative fluorescence PCR. No exogenous viral contamination was detected in the prepared RM. Homogeneity verification showed no significant intra-assay and inter-assay differences, indicating excellent homogeneity. Stability tests demonstrated that the RM could be stably stored at -20 \u2103 for 6 months, 4 \u2103 for 7 days, and 25 \u2103 for 24 hours. The certified concentration of the ORF101 gene of the RM was (8.6 \u00b1 1.3) \u00d7 10\u00b3 copies\/\u03bcL, which was applicable to quantitative fluorescence PCR detection. In conclusion, the developed RM with accurate quantification and favorable homogeneity can be used for quality control, instrument calibration, and methodological evaluation of LSDV nucleic acid detection. It also provides a reliable material basis for the research and development of in vitro diagnostic reagents for LSDV.<\/p>\n\n\n\n
