\u4f20\u67d3\u6027\u76ae\u4e0b\u53ca\u9020\u8840\u7ec4\u7ec7\u574f\u6b7b\u75c5\u6bd2\uff08IHHNV\uff09\u611f\u67d3\u5bf9\u867e\u5f15\u53d1\u6025\u6027\u4f20\u67d3\u75c5\uff0c\u5bf9\u5e7c\u867e\u5371\u5bb3\u5c24\u4e3a\u663e\u8457\u3002\u4f9d\u636e Genbank \u6536\u5f55\u7684 IHHNV \u57fa\u56e0\u5e8f\u5217\uff08AF218266.2\uff09\uff0c\u5236\u5907\u542b IHHNV \u5168\u57fa\u56e0\u7ec4\u5e8f\u5217\u7684\u5047\u75c5\u6bd2\u6807\u51c6\u7269\u8d28\uff08NIM-RM4062\uff09\u3002\u9488\u5bf9 ORF1 \u57fa\u56e0\u8bbe\u8ba1\u7279\u5f02\u6027\u5f15\u7269\u63a2\u9488\uff0c\u6784\u5efa IHHNV \u6570\u5b57 PCR \u68c0\u6d4b\u65b9\u6cd5\uff0c\u5b8c\u6210\u5047\u75c5\u6bd2\u6838\u9178\u6807\u51c6\u7269\u8d28\u7684\u5747\u5300\u6027\u4e0e\u7a33\u5b9a\u6027\u8003\u5bdf\u3002\u8be5\u6807\u51c6\u7269\u8d28\u62f7\u8d1d\u6570\u6d53\u5ea6\u6807\u51c6\u503c\u53ca\u6269\u5c55\u4e0d\u786e\u5b9a\u5ea6\uff1a(1.22 \u00b1 0.15) \u00d7 10\u00b3 copies\/\u03bcL (k=2)\u3002\u591a\u5e73\u53f0\u6570\u5b57 PCR \u5b9a\u503c\u9a8c\u8bc1\u663e\u793a\uff0c\u76f8\u5bf9\u6807\u51c6\u504f\u5dee\u5c0f\u4e8e4 %\uff0c\u91cf\u503c\u8de8\u5e73\u53f0\u590d\u73b0\u6027\u826f\u597d\u3002\u7ecf\u5546\u7528\u6838\u9178\u68c0\u6d4b\u8bd5\u5242\u76d2\u6bd4\u5bf9\uff0c\u5176\u91cf\u503c\u4e0e\u8367\u5149\u5b9a\u91cf PCR \u68c0\u6d4b\u7ed3\u679c\u4e00\u81f4\u6027\u4f73\u3002<\/p>\n\n\n\n

Infectious hypodermal and haematopoietic necrosis virus (IHHNV) infection causes acute infectious diseases, which severely endangers juvenile shrimp. According to the IHHNV gene sequence (AF218266.2) deposited in GenBank, pseudovirus reference material carrying the full-length IHHNV genome was prepared. Specific primers and probes targeting the ORF1 gene were designed to establish a digital PCR detection assay. The uniformity and stability of the reference material were verified via digital PCR. The certified value and expanded uncertainty were (1.22 \u00b1 0.15) \u00d7 10\u00b3 copies\/\u03bcL (k=2). Validation across multiple dPCR platforms presented relative standard deviations lower than 4 %, indicating favorable reproducibility. Detection results obtained by commercial IHHNV nucleic acid kits showed great consistency with the certified values.<\/p>\n\n\n\n
