\u4e3a\u5b9e\u73b0\u5c0f\u53cd\u520d\u517d\u75ab\u75c5\u6bd2\uff08Peste des petits ruminants virus, PPRV\uff09\u7684\u5feb\u901f\u3001\u51c6\u786e\u3001\u5b9a\u91cf\u68c0\u6d4b\uff0c\u4ee5\u56fd\u5185 PPRV Clone 9 \u75ab\u82d7\u682aN<\/em>\u57fa\u56e0\u4e3a\u9776\u4f4d\u70b9\u8bbe\u8ba1\u7279\u5f02\u6027\u5f15\u7269\u53ca\u63a2\u9488\uff0c\u5efa\u7acb\u6db2\u6ef4\u5f0f\u6570\u5b57 PCR\uff08Droplet digital PCR, ddPCR\uff09\u65b9\u6cd5\uff0c\u5e76\u8bc4\u4ef7\u5176\u7279\u5f02\u6027\u3001\u654f\u611f\u6027\u53ca\u91cd\u590d\u6027\u3002\u5bf9\u6bd4\u8be5\u65b9\u6cd5\u4e0e\u5b9e\u65f6\u8367\u5149\u5b9a\u91cf PCR\uff08Fluorescence quantitative PCR, qPCR\uff09\u7684\u7075\u654f\u5ea6\uff0c\u540c\u65f6\u7528\u4e8e PPRV\uff08Clone 9 \u682a\uff09\u6838\u9178\u6807\u51c6\u7269\u8d28\u5b9a\u91cf\u68c0\u6d4b\u3002\u7ed3\u679c\u663e\u793a\uff0c\u8be5 ddPCR \u65b9\u6cd5\u7279\u5f02\u6027\u826f\u597d\uff0c\u4ec5 PPRV \u68c0\u6d4b\u5448\u9633\u6027\uff0c\u5176\u4f59\u7f8a\u6e90\u75c5\u6bd2\u5236\u54c1\u53ca\u6bd2\u682a\u68c0\u6d4b\u5747\u4e3a\u9634\u6027\uff1b\u68c0\u6d4b\u7075\u654f\u5ea6\u4f18\u5f02\uff0c\u6700\u4f4e\u68c0\u51fa\u9650\u4e3a4.33 \u62f7\u8d1d \/\u03bcL\uff0c\u76f8\u8f83 qPCR \u65b9\u6cd557.3 \u62f7\u8d1d \/\u03bcL\u7684\u68c0\u51fa\u9650\uff0c\u7075\u654f\u5ea6\u9ad8\u51fa 10 \u500d\uff1b\u91cd\u590d\u6027\u53ef\u9760\uff0c\u8bd5\u9a8c\u53d8\u5f02\u7cfb\u6570\u4e3a1.8 %\uff1b\u5b9a\u91cf\u7ed3\u679c\u7cbe\u51c6\uff0c9 \u5bb6\u5177\u5907\u8d44\u8d28\u5355\u4f4d\u7684\u68c0\u6d4b\u6570\u636e\u7ec4\u95f4\u53d8\u5f02\u7cfb\u6570\u5747\u5c0f\u4e8e5 %\u3002\u672c\u65b9\u6cd5\u53ef\u9ad8\u6548\u5b8c\u6210 PPRV \u6838\u9178\u5feb\u901f\u68c0\u6d4b\uff0c\u53ef\u4e3a\u8be5\u75c5\u6bd2\u4e34\u5e8a\u8bca\u65ad\u4e0e\u6838\u9178\u7edd\u5bf9\u5b9a\u91cf\u63d0\u4f9b\u53ef\u9760\u6280\u672f\u624b\u6bb5\u3002<\/p>\n\n\n\n

In order to achieve rapid, accurate and quantitative detection of Peste des petits ruminants virus (PPRV), a droplet digital PCR (ddPCR) method was established with specific primers and probe targeting the N<\/em> gene of the domestic vaccine strain PPRV Clone 9, and the specificity, sensitivity and repeatability of the method were evaluated. At the same time, the sensitivity of the established ddPCR method was compared with that of the real-time fluorescence quantitative PCR (qPCR) method. Finally, the established ddPCR method was applied to the quantification of PPRV (Clone 9 strain) ribonucleic acid reference material. The results showed that the ddPCR method had good specificity, with only positive results for PPRV detection and negative results for other sheep-derived virus biological products and viruses. The sensitivity of the ddPCR method was high, with a detection limit of 4.33 copies\/\u03bcL, which was 10 times higher than the sensitivity of the qPCR method (57.3 copies\/\u03bcL). The coefficient of variation of the repeatability test was 1.8 % in the ddPCR method. Besides, the ddPCR method was accurate in quantification, as the coefficient of variation was less than 5 % between groups of measurement data in the nine qualified institutions. The ddPCR method established in this study can effectively and rapidly detect PPRV nucleic acid, providing an effective method for the diagnosis and absolute quantification of PPRV.<\/p>\n\n\n\n
